The Effect of Antisperm Globulin on Spermatogenesis in Rats

Abstract
Antisperm globulin.(A.S.G) produced by immunication of female rabbit with emulsion of epididymal sperm cells of the white rat and complete Freund¡¯s adjuvant was injected into normal white male rats every day for 5 days and the effect of antisperm globulin on spermatogenesis and mature spermatozoa in the treated (passively immunized) rats was observed at the light microscopic and electron microscopic levels. The treated male rats and the non-treated control animals were mated with sexually active normal female rats and impregnation was observed, respectively, and the following results were obtained: 1. On the light microscopic examination of the testis and the epididymis of the rats 5 days after treatment with A.S.G. the seminiferous tubules showed an impairment of spermatogenesis. The epithelial cells of the tubules became atropic and thinner and spermatozoa in the lumne were few. The epididymal epithelium was intact but the intraluminal spermatozoa were markedly decreased in number, being agglutinated each other. 2. Electron microscopic finding of the testis one year after treatment with A.S.G. included destructive changes in the acrosomes of spermatozoa and spermatids with irregular membranes of the nuclei and dilation of endoplasmic reticulum. The swelling and decrease in number of cristae at the mitochondria were also seen in the germinal cells. 3. Impregnation occurred in none of 10 treated rats while all of 10 treated controls impregnated their mates.
